Depending on the library prep chemistry, mechanical shearing using Covaris g-TUBEs is an optional step during the Ligation library prep, while enzymatic shearing is an integral step in our Rapid approach, see the technical document for more details

Alternative mechanical shearing methods to gTUBEs are possible, such as sonication, acoustic shearing, hydrodynamic shear, and needle shearing.

The validated approaches are summarized here. If other approaches are considered, optimization by the end user will be required.
