Our Direct RNA (SQK-RNA002) and cDNA (SQK-PCS111 and SQK-PCB111) kits are designed to target 3’-polyadenylated transcripts for sequencing.

If the RNA of interest is not polyadenylated, a third-party polyA polymerase, such as this kit, can be used prior to library prep.

Alternatively, total RNA can be used as input at the user’s discretion, provided enough total RNA is used such that the fraction of poly-adenylated material is sufficient to meet the input requirements for the kit being used.
