What’s the purpose of the dephosphorylation of the genomic DNA in Cas9 targeted sequencing?

We highly recommend performing dephosphorylation of the genomic DNA step (~15 min) to remove the 5’ phosphates from the ends of the genomic DNA.

This step will help reducing the ligation of sequencing adapters to non-target DNA therefore background reads will be heavily reduced.
