Pore occupancy is the ratio of pores in 'strand' to 'single pore'. A good pore occupancy would be \>75%.
There are several reasons that could lead to low pore occupancy, though usually, it is because you do not have enough successfully adapted library in the final stage.
This is usually due to either:
- Inefficient bead clean-ups
- Inefficient adapter ligation
For both of the above reasons, we advise waiting longer during both clean-up and ligation steps.
