If you are planning on storing a prepared library, please do before the "Priming and loading the SpotON flow cell" step of the protocol.

Do not add the SQB and LB until immediately before loading into a flow cell. You can store the library in Eppendorf LoBind tubes at 4 °C for short-term or repeated use, or at -80 °C for single use or longer-term storage of more than 3 months.

The DNA Library Stability know-how contains more details and benchmarks with the Ligation kit (SQK-LSK109). If you have already added the SQB and LB, the library must be used immediately.
